01 Top discoveries — evidence graded
| Compound | Grade | Key role |
| C-H-O-N (base organic) | A | L-DOPA — the gold-standard PD drug — belongs to this class |
| C-H-O-N-S (sulfur amino acids) | A (human PD trials) | NAC increased dopamine-transporter binding on DaTscan; GSH depletion is an early PD marker |
| C-H-O-N-Mg (magnesium) | B+ | NMDA antagonism; muscle-rigidity support; PD risk association |
| C-H-O-N-S-Zn (zinc-cysteine) | B+ | Metallothionein neuroprotection; MT-I/II protect against MPTP |
| C-H-O-N-Zn-F/I/Br/Cl (zinc-halogens) | NOVEL — zero data | Genuinely original prediction; requires laboratory investigation; excluded from the formula |
| C-H-O-N-Fe (iron) | A (ID) / harmful | Tyrosine-hydroxylase cofactor BUT accumulation drives ferroptosis — excluded |
02 Strongest evidence — NAC / glutathione (human PD data)
- Monti et al. 2019 (PMID 31048906) — DaTscan SPECT showed significant increases in dopamine-transporter binding (caudate & putamen) after NAC.
- Coles et al. 2018 (PMID 29304084) — MR spectroscopy confirmed NAC increases brain glutathione in PD patients.
- Monti et al. 2016 (PMID 27708942) — NAC improved blood antioxidant measures (GSH/GSSG).
- Sian 1994 (PMID 8210224) — GSH depletion in the substantia nigra is one of the earliest detectable PD changes.
03 Novel discovery & metal context
- Zinc-halogen amino acids (Zn-F / Zn-I / Zn-Br / Zn-Cl): unique to the Parkinson’s output, with zero published research on these classes in neurodegeneration. Excluded from the practical formula pending safety/efficacy research.
- Iron and copper are dopamine-synthesis cofactors but accumulate pathologically in the PD substantia nigra — iron is EXCLUDED.
- Zinc is kept LOW (15 mg) to support BDNF while minimizing alpha-synuclein aggregation risk.
04 What is in the package
- Discovery data (JSON) — compounds with probability scores and pathways.
- Known-properties report (peer-review corrected, evidence-graded).
- 30-day formula with pharmacist compounding specs (CAS numbers, USP-grade forms, interactions).
- Patent application + four patent drawings (SVG); USPTO forms list.
- Discovery script for reproducing and extending the research.
